After Peter gains his superpower in the first part of the movie, he uses the power on Flash Thompson to humiliate him as he is prone to bully Peter frequently. But the principal of his school calls Uncle Ben, and he asks Peter to walk his aunt home for him that night which Peter forgets to do so as he was distracted with helping Dr. Curt Connors. This makes Uncle Ben even angry. He talks to Peter about his father’s philosophy of responsibility, which leads to Peter storming out and being hurt to remind him of his father, who abandoned him. Uncle Ben follows after Peter. Peter goes to a grocery store, where a thief robs it, and Peter doesn’t try to stop him to spite the cashier. Out near the Grocery Store, the thief is confronted by Uncle Ben while the thief drops the gun. At the same time, Uncle Ben and the thief struggle for the weapon until Ben is shot on the torse by the thief and is left to die.

Be it a comic or Tv Series, Movie, Uncle Ben was killed by a vigilante on the run who was either trying to steal from him or robbing their house. And for most of it, Peter has always had an encounter with the vigilante, who he failed to stop.
